CE–MS Approaches for Glyco(proteo)mic Analysis

Abstract

Glycans, also known as carbohydrates or saccharides, are compounds that consist of several monosaccharides and can be present in their free form or complexed with another molecule. Glycoconjugates can be divided into three distinct groups: glycoproteins, glycolipids, and glycosaminoglycans. This chapter focuses on the analysis of glycoproteins. It provides a broad overview of the various applications where capillary electrophoresis–mass spectrometry (CE–MS) is being utilized in a glyco(proteo)mic setting, being it on released glycan, glycopeptide, or intact level. Glycans can be attached to a protein via the nitrogen atom of an asparagine residue, generating the well-known N -linked glycosylation. The chapter discusses the general applications for glycomics analysis with a main focus on N -linked glycans, providing insights into various glycan release strategies, derivatization methods, and N -glycan analysis by CE–MS in either positive or negative mode.
